We found that the expression of DVRM was up-regulated in the early stage of osteogenesis, which is consistent with the fact that DVRM is a positive regulator of osteoblast differentiation. It has been reported that EZH has multiple functions in cell proliferation, migration, and apoptosis.
In addition, it can inhibit cancer cell growth through regulating cell cycle, DNA repair and apoptosis ([@B23],[@B24]). Here, we found that EZH was significantly down-regulated during osteogenesis, which is consistent with the fact that EZH acts as a negative regulator of osteogenic differentiation. Furthermore, we also observed that EZH can promote osteoblast proliferation by repressing DVRM. In summary, our results showed that EZH can promote osteogenic differentiation of hBMSCs by suppressing DVRM.
Our study has several limitations. First, we did not investigate whether the expression of EZH is changed in human bone marrow mesenchymal stem cells (hBMSCs) at different stages of osteogenesis, which could be used to further elucidate the role of EZH in the process of osteogenesis. Second, the detailed mechanism of how EZH regulates DVRM remains unclear. Although we demonstrated that EZH promoted osteogenesis through suppressing DVRM, more experiments should be done to clarify the mechanism.
